Glajch: The Role of Multi-Dimensional Separations and Mass Spectrometry for the Development of Complex Generics and Biologics

Abstract:

The development of certain complex pharmaceutical drugs poses unique challenges for the analytical chemist. For example, a subset of current drugs does not contain a single, unique chemical structure, but rather mixtures of many related compounds. Examples of such drugs include low molecular weight heparins, polypeptide mixtures, and many biotherapeutic protein products, which may comprise molecules with different forms of glycosylation, protein sequence variations, and/or side chain modifications.

The role of multi-dimensional separations, mass spectrometry, and other complementary techniques will be examined in the development of complex generics and biologics. This will include the overall approach to support equivalence of different lots of materials, the use of SEC-MALS for characterization of heparins, and multi-dimensional LC/MS/MS for the analysis of an enzymatic digest of a polypeptide mixture.

About the Speaker:

Joseph Glajch is currently Director of Analytical Development at Momenta Pharmaceuticals (Cambridge, MA). His research interests include HPLC method development, column characterization and development, and the application of separations technology to chemical and pharmaceutical development. Dr. Glajch received an AB degree in Chemistry at Cornell and a PhD in Analytical Chemistry from the University of Georgia under Prof. L. B. (Buck) Rogers and then joined the Central R&D Department at the DuPont Experimental Station in Wilmington, DE. He has held various scientific and management positions at DuPont and later as part of BMS Medical Imaging in Billerica, MA. He has co-authored more than 40 scientific papers, 3 books, and 5 patents and served on the editorial board of Analytical Chemistry, the Journal of Chromatography, and LC/GC. He also served as Chairman of the American Chemical Society Division of Analytical Chemistry and the Chromatography Forum of the Delaware Valley and was the recipient of the 1994 Chromatography Forum on the Delaware Valley
Award.
