Mixed-Mode Chromatography in the Purification of Recombinant Proteins

Abstract:

Mixed-mode chromatography sorbents, which combine multiple interaction mechanisms in a single chromatography step, offer unique selectivity and a reduction in the number of process steps. The mixed-mode properties of the sorbents increase the number of experimental variables to be evaluated but an understanding of the resin properties facilitates method development. We show the use of mixed mode sorbents in the purification of E. coli recombinant proteins, with applications for capture, intermediate purification and polishing of recombinant proteins, as well as how they can be used to eliminate even massive amounts of LPS.

About the Speaker:

Andrew Lees is currently Scientific Director of Fina BioSolutions, a consulting laboratory in Rockville, Maryland. The company provides protein purification and bioconjugation services. He was formerly Director of Macromolecular Sciences at Biosynexus, Inc. Andrew holds 15 patents, mainly in his specialty area of protein-polysaccharide conjugate vaccines. He is also an adjunct at the Uniformed Services University of the Health Sciences in Bethesda, Maryland and is a Senior Scientist at the University of Maryland. He received his Ph.D. in biophysics from the Johns Hopkins University and an undergraduate degree in chemistry from Harvey Mudd College.
