About This Meeting

Analysis of proteins and metabolites by mass spectrometry has made extraordinary advances in the past 10 years, however improvements to the number of molecules measured has not led to equivalent improvements in translated biomarkers to the clinic.  We believe that translation of human proteomics and metabolomics studies to validated biomarkers has been limited in general by poor experimental design at the earliest stages of discovery. Some typical limitations include low sample numbers and generally a lack of quality control (QC) along the discovery pipeline, including the steps of sample preparation, analytical data collection, and data analysis.  In metabolomics, more attention has been paid to the number of features measured as opposed compound identity or analytical quantification. Our lab has focused on using our own mistakes along with first principles of analytical chemistry to revisit the biomarker pipeline and subsequently improve the quality of these discovery experiments. We have undertaken this with the goal of performing biomarker discovery in clinically relevant sample cohorts with methods that are more easily translatable to a clinical laboratory, hopefully to decrease the rate of biomarker attrition along the translational pipeline.  This presentation will use examples from the areas of hepatitis/liver fibrosis, cardiovascular disease, and sepsis to describe important sample preparation, separation, and mass spectrometry workflows and also pitfalls along the biomarker pipeline.
